Transfection is the process by which nucleic acids are introduced into eukaryotic cells. (…) Transfection efficiency is a critical parameter to increase biologics’ productivity; the right protocol has to be identified to ensure high transfection efficiency and therefore high product yield. Design of experiments (DoE) is a mathematical method that has become a key tool in bioprocess development. Based on the DoE method, we developed an operational flow that we called “Design of Transfections” (DoT) for specific transfection modeling and identification of the optimal transfection conditions.
La trasfezione è il processo mediante il quale gli acidi nucleici vengono introdotti nelle cellule eucariotiche. (…) L’efficienza della trasfezione è un parametro critico per aumentare la produttività dei biologici; è necessario identificare il protocollo giusto per garantire un’elevata efficienza di trasfezione e quindi un’elevata resa del prodotto. Il disegno sperimentale – Design of Experiment (DoE) – è un metodo statistico che è diventato uno strumento chiave nello sviluppo dei bioprocessi. Sulla base del DoE, abbiamo sviluppato un flusso operativo che abbiamo chiamato “Design of Transfections” (DoT) per la modellazione specifica della trasfezione e l’identificazione delle condizioni ottimali di trasfezione.
